T‐cell mediated responses against alpha‐foetoprotein in hepatocellular carcinoma: Relationship with hepatitis C virus infection, tumour phenotype and patients’ survival

چکیده
Background Alpha-foetoprotein (AFP) is a potential immunotherapeutic target in hepatocellular carcinoma (HCC). However, T-cell response (TR) to AFP suppressed HCC due immune evasion. It unknown whether HCV infection may pre-condition TR against AFP, or influence the clinical course of HCC. Methods We prospectively enrolled 18 HCV+ treatment-naïve patients with cirrhosis (CC), cases and 17 HCV- cases. was quantified by ELISPOT using assays specific interleukin (IL) 2, IL10 granulocyte-monocyte colony stimulating factor (GM-CSF) on ex-vivo peripheral blood mononuclear cells (PBMC) stimulated vitro peptides. Cytokine ratios were compared between groups clinicopathological features HCC, including overall survival (OS). Results The proportion AFP-specific responses not different across studied for any assayed cytokines. IL-2 increased larger (P = .02), multifocal tumours .01) correlated advanced disease .01). TRs did correlate other factors predict OS. Conclusion Tumour stage but related emergence anti-AFP TRs. These data enable formulation rationale further development immunotherapy facilitating optimal patient selection future studies.

The Absence of Hepatitis C Virus Infection Among Patients with Hepatitis B virus in Mashhad, Iran
Background and Aims: Many studies have provided evidence for the role of hepatitis B and C viruses in the development of liver cancer. Although the routine treatment is available for both conditions, no definite guideline is available to treat patients dually infected with HBV and HCV. This study was performed to determine the frequency of HBV/HCV-coinfection in Mashhad, North-East of Iran. Mat...

ABSTRACT Background: Hepatitis B virus (HBV) infection in patients who lack detectable hepatitis B surface antigen (HBsAg) is called occult hepatitis B infection. Such infections have been frequently identified in patients with chronic hepatitis Cliver disease, but their prevalence is not known. Methods: 207 patients with chronic hepatitis C who were HCV -RNA and antiHCV positive were stu...
متن کاملChronic hepatitis C virus infection and pathogenesis of hepatocellular carcinoma.
Hepatitis C virus (HCV) infection is one of the major causes of advanced liver disease and hepatocellular carcinoma (HCC) worldwide. While the knowledge about the molecular virology of HCV infection has markedly advanced, the molecular mechanisms of disease progression leading to fibrosis, cirrhosis and HCC are still unclear. Accumulating experimental and clinical studies indicate that HCV may ...
متن کاملStatins and the risk of hepatocellular carcinoma in patients with hepatitis C virus infection.
PURPOSE Statins may have protective effects against cancer, but no studies have focused on their effects in patients with chronic hepatitis C virus (HCV) infection. The purpose of this study was to investigate the association between use of statins and risk of hepatocellular carcinoma (HCC) in HCV-infected patients. PATIENTS AND METHODS Ours was a population-based cohort study of 260,864 HCV-...

Hepatocellular carcinoma in woodchuck were characterized for woodchuck hepatitis virus integration nea c-myc oncogene. In one tumor, viral integration resulted in overexpression of a c-myc viral cotranscript. In a second tumor, viral insertion, 600 bp upstream of c-myc exon 1, was associated with increased levels of normal c-myc mRNA. These results demonstrate that integration of woodchuck hepa...
